DEPT谱的特点：
1、分辨能力高 ：谱线之间分得很开，容易识别。
2、化学位移范围大：0~300 ppm，是NMR谱的20~30倍 。
3、掌握碳原子，特别是无H连接时的信息，确定碳原子级数。
4、容易实现双共振实验。
5、准确测定驰豫时间，可作为化合物结构鉴定的波谱参数， 帮助指认碳原子。
6、虽然不能直接观察到季碳，但结合全图可准确识别季碳，且DEPT谱碳数更少，图谱更清晰。可以克服偏共振试验中共振谱线发生重叠和指配复杂分子的谱图时的一些困难。测量时间比偏共振谱短。

DEPT谱是在NMR中用来区分伯仲叔季碳的一种谱图。信号强度仅仅与脉冲倾倒角有关，
为了区分不同的碳，一般要做三次实验，分别为不同的角度，其中季碳不出峰：
脉冲倾倒角为135度的DEPT谱图：甲基，次甲基的峰向上（即信号为正），亚甲基为倒峰（即信号为负）
脉冲倾倒角为90度的DEPT谱图：只能看到次甲基向上的峰
脉冲倾倒角为45度的DEPT谱图：所有的次甲基、亚甲基、甲基的峰都向上（不常用，因为无法达到区分的目的）
通过135度和90度谱图即可区分出伯碳、仲碳、叔碳，由于季碳在所有的DEPT谱图中都没有信号，因此只要和全谱比较，就很容易的得到季碳。
